Tim-3在宫颈癌组织中的表达及其与恶性进展的关系

Relation of Tim-3 Expression in Cervical Cancer to Tumor Progression

  • 摘要: 目的 探讨宫颈癌细胞表达T-cell immunoglobulin and mucin domain containing 3 protein(Tim-3)与宫颈癌恶性进展的关系。方法 用反转录PCR(RT-PCR)、Western blot和共聚焦显微镜检测宫颈癌细胞系HeLa和SiHa中Tim-3基因和蛋白的表达及其胞内定位;用免疫组织化学法检测43例宫颈癌组织中Tim-3的表达,并分析其与临床分期、组织分型、转移等相关性。结果证实HeLa和SiHa两个宫颈癌细胞系均表达Tim-3,在前者Tim-3分布于胞质,而在后者则分布于核内及其周围。在宫颈癌患者病理标本中可见癌巢和肿瘤血管高表达Tim-3,且Tim-3表达强度与与临床分期、组织分型和转移相关。结论宫颈癌细胞及血管内皮细胞均高表达Tim-3,且Tim-3表达强度与宫颈癌恶性进展及转移高度相关。

     

    Abstract: Objective To confirm the relationship between Tim-3 (T-cell immunoglobulin and mucin domain containing 3 protein) expression in cervical cancer and the progression of the tumor. Methods RT-PCR,Western blot and confocal microscopy were used to detect the expression of Tim-3 in cervical cancer cell lines,HeLa and SiHa,at both gene and protein levels and its intracellular distribution.Meanwhile,specimens from 43 patients with cervical cancer were detected the expression of Tim-3 by immunohistochemistry.We analysed the relation of Tim-3 expression to clinical stage,tissue typing,metastasis. Results Tim-3 was expressed highly in the HeLa and SiHa cells.Moreover,according to the statistics results,Tim-3 expressed highly in the cancer cell,relative to the increased degree of malignancy of cervical cancer. Conclusion Tim-3 is expressed highly in cervical cancer cell and vessel endothelial cell,moreover it is relative to the increased degree of malignancy of cervical cancer.
