皮肤瘢痕和瘢痕鳞状细胞癌中Cx26基因表达及突变的意义

Significance of Cx26 Gene Expressions and Mutation in Skin Scar and Skin Scar Squamous Cell Carcinoma

  • 摘要: 目的探讨Cx26基因表达及突变在皮肤瘢痕及瘢痕鳞状细胞癌(以下简称瘢痕鳞癌)中的意义。方法采用免疫组织化学SP法及原位杂交技术检测15例皮肤瘢痕和22例瘢痕鳞癌中Cx26蛋白及mRNA的表达。分别从14例皮肤瘢痕和瘢痕鳞癌石蜡包埋组织中提取DNA,运用PCR扩增及测序的方法进行突变筛查。利用计算机图像分析系统采集、分析图像,并对数据进行统计学处理。结果(1) Cx26蛋白、Cx26mRNA在正常皮肤表皮中阳性,在皮肤瘢痕上皮呈强阳性表达,在瘢痕鳞癌组织中呈弱阳性或阴性表达。正常皮肤组、皮肤瘢痕组及瘢痕鳞癌组两两比较,其表达(阳性面积和平均吸光度)差异均有统计学意义(P<0.05)。(2)测序后未发现瘢痕鳞癌中有Cx26基因突变。结论Cx26蛋白及其mRNA的高表达,对瘢痕上皮增生具有促进作用;其表达下降与瘢痕鳞癌的发生有关;基因突变位点有待进一步探讨。

     

    Abstract: ObjectiveTo investigate the significance of Cx26 gene expression and mutation in skin scar and skin scar squamous cell carcinoma. Methods The expressions of Cx26 protein and its mRNA in 15 skin scar and 22 skin scar squamous cell carcinomas were detected by immunohistochemical method of SP and in situ hybridization.To analyze the characteristics of Cx26 gene mutations,DNA was extracted from 28 samples(14 skin scar and 14 skin scar squamous cell carcinoma tissues)of Paraffin embedded and screened for mutation by direct sequencing.Pictures were collected and analyzed,statistical analysis was performed by SPSS system. Results (1)Cx26 and its mRNA showed positive expression in normal skin epidermis and strong positive in pathological skin scar epithelium,weakly positive or negative expression in skin scar squamous cell carcinoma tissues.Each of two groups was statistically different(P<0.05).(2)The Cx26 genes were not examined to have the mutation in skin scar squamous cell carcinoma tissues after sequencing.ConclusionThe high expression of Cx26 protein and Cx26mRNA may be related to the hyperplasia of skin scar epithelium.The low expression may be related to the occurrence of skin scar squamous cell carcinoma.The genetic mutation suits need more explored.
