Role of CT Scan,SOD and MDA Expression in Radiation-induced Lung Injury

  • 摘要: 目的通过对照射后的大鼠肺部进行CT扫描和血清中SOD、MDA的检测,探讨它们在放射性肺损伤中的预测价值。方法SD大鼠分为3组:对照组、7.0 Gy组、14.4 Gy组。大鼠麻醉后用60Co治疗机单次照射右全肺;在照射后30天、90天对大鼠肺部进行CT扫描,并于照射后1、7、30、90 d处死各组大鼠,取大鼠右肺组织进行HE、Masson 染色,并对血清中SOD、MDA的含量进行检测。结果对照组、7.0 Gy组大鼠右肺组织未见明显的组织病理学改变,而14.4 Gy组可见明显纤维化改变。CT扫描结果:对照组、7.0 Gy组均未见明显的实变影;而14.4 Gy组,照射野内可见高密度实变影。7.0 Gy组大鼠SOD与 MDA水平与对照组比较差异无统计学意义;14.4 Gy组大鼠SOD水平在照射后1 d明显升高,30、90 d时则明显降低,但与对照组比较差异无统计学意义,而MDA水平从第1天开始即明显升高,且持续维持在较高的水平,各时间点与对照组比较差异均有统计学意义(P<0.05)。结论超过“阈值”的剂量照射大鼠肺组织后,CT扫描有一定的诊断价值,但其预测作用是滞后的;未发现SOD有明确的预测价值;初步认为MDA对放射性肺损伤有一定的预测价值。

     

    Abstract: ObjectiveTo evaluate the role of CT scan,SOD and MDA expression in radiation lung injury. Methods The right hemithorax of SD rats were irradiated with a single dose of 7.0 Gy and 14.4 Gy. All animals were sacrifaced and the lung were removed at 1,7,30,90 days after radiation. The specimens were stained with HE,Masson to observe the histopathological changes.And the expression of SOD and MDA in serum by ELISA. ResultsThere were no notable hispathological changes in control groups and 7.0 Gy group. Otherwise,the formation of fibrosis could be observed in 14.4 Gy group. CT scan showed no significant opacities in the control group and 7.0 Gy group. However,there were scattered patchy of high intensity in the radiation field in 14.4 Gy group after radiation. Compared with the control group,the expression level of SOD and MDA had no significant difference in 7.0 Gy group at all time points. After 14.4 Gy exposed,The level of SOD was increased at 1 day and decreased at 30,90 days. The level of MDA was significantly elevated at 1day and remained high level at each time point. ConclusionMore than “threshold” dose of radiation,the image changes had occur in lung tissue,there had some clinical value,but the predictive effect of CT scanning was delayed,because image changes often occur after pathological physiological changes. Compared with SOD,the expression of MDA not only could be tested in blood but also was relatively stable which showed that it played an important role to predict the radiation-induced lung injury.
