白藜芦醇对小鼠Lewis肺癌细胞生长的抑制作用及其机制

Inhibitory Effects of Resveratrol on Growth of Lewis Lung Cancer Cell in Mice and Possible Mechanism

  • 摘要: 目的探讨白藜芦醇(Res)对小鼠Lewis肺癌的抑瘤效应及其作用机制。方法将32只接种Lewis肺癌细胞的C57BL小鼠随机分为4组:对照组、Res低剂量组、Res高剂量组和顺铂组(阳性对照),每组8只,用药干预14天,于接种后第20天起处死,剥离皮下瘤,称瘤重,计算抑瘤率,通过Western blot检测移植瘤巨噬细胞移动抑制因子 (MIF)表达,并用原位末端标记法检测肿瘤细胞凋亡指数。结果Res低剂量组、Res高剂量组和顺铂组抑瘤率分别为11.54%、54.58%、62.32%,Res高剂量组和顺铂组瘤重显著低于对照组(P<0.01);Res高剂量组和顺铂组较对照组MIF蛋白表达水平降低,而凋亡指数增加,差异均有统计学意义(P<0.01)。结论白藜芦醇具有抑制小鼠Lewis肺癌生长的作用,其机制可能与抑制MIF蛋白表达和诱导肿瘤细胞凋亡有关。

     

    Abstract: ObjectiveTo explore the inhibitory effects of resveratrol(Res) on growth of Lewis lung cells in mice and the underlying mechanism. MethodsThirty-two C57BL mice inoculated Lewis lung cancer cell were randomly divided into 4 groups (each group with 8 mice): control group, low-dose Res group, high-dose Res group and cisplatin group(positive control). Different treatments were administrated from day 4 to day 17 after inoculation, then all mice were sacrificed. The tumors were removed and weighed to calculate inhibitory rate. Expression of macrophage migration inhibitory factor(MIF) was detected by western blot. Apoptotic index of tumor cells was determined by TUNEL assay. ResultsThe inhibitory rates were 11.54%, 54.58% and 62.32% in low-dose Res group, high-dose Res group and cisplatin group respectively. Tumor weight and levels of MIF protein expression were significantly lower in high-dose Res group and cisplatin group compared with control group(P<0.01) . Apoptotic index was enhanced in high-dose Res group and cisplatin group than that in control group(P<0.01). Conclusion Res had a strong inhibitory effects on growth of Lewis lung cancer by inhibition of MIF protein expression and induction of tumor cell apoptosis.
