四物合剂对马利兰诱导HL-60细胞凋亡的 影响

Effect of Siwu Mixture on Apoptosis Induced by Myleran in HL-60 Leukemic Cells

  • 摘要: 目的观察不同剂量四物合剂对马利兰诱导HL-60细胞凋亡作用的影响。方法MTT法检测细胞增殖活性,流式细胞术检测细胞凋亡情况,Western blot检测细胞p53/bcl-2表达,ELISA法检测细胞色素C释放,分光光度法检测Caspase-3、Caspase-8和谷胱甘肽(GSH)。结果四物合剂可激活细胞Caspase-3和Caspase-8,上调细胞p53/bcl-2表达比率、升高细胞色素C释放、降低GSH(P<0.05,P<0.01),且60mg/L剂量可抑制HL-60细胞生长(P<0.05)。结论四物合剂可抑制HL-60细胞生长,对马利兰诱导HL-60 细胞凋亡具有增效作用,其增效作用机制可能与激活肿瘤细胞凋亡PKC通路有关。

     

    Abstract: ObjectiveTo observant the dose effects of Siwu Mixture on the apoptosis in HL-60 leukemic cells by myleran. MethodsHL-60 leukemic cells proliferation was tested by MTT assay,cell apoptosis was analyzed by flow cytometry,the p53/bcl-2 expression was assessed by Western blot, cytochrome C release was determined by ELISA,GSH,Caspase-3 and Caspase-8 activation were examined by colorrimetric assay. ResultsSiwu mixture could increase HL-60 leukemic cell apoptosis by myleran,upregulated ratio of p53/bcl-2 expression,activated of Caspase-3 and Caspase-8,increased the release of cytochrome C,decreased the levels of GSH(P<0.05,P<0.01),and inhibited the growth of HL-60 leukemic cells at the dose of 60mg/L(P<0.05). ConclusionSiwu mixture was able to inhibit the growth of HL-60 leukemic cells,enhance antitumor effect of myleran and the major mechanism siwu mixture's promotion sensitivity of tumor cell to chemotherapeutic agents might be related to activat apoptosis signal pathway.
