hMLH1、转化生长因子β受体Ⅱ在食管癌中的表达及意义

Expression and Significance of hMLH1 and TGFβRⅡ in Esophageal Squamous Cell Carcinoma

  • 摘要: 目的:探讨hMLH1、转化生长因子β受体Ⅱ在食管黏膜上皮癌变过程中的意义及相关性。方法采用免疫组织化学方法,检测正常食管黏膜上皮及食管不同病变组织中hMLH1、转化生长因子β受体Ⅱ的表达情况, 并分析其与临床病理参数的相关性。结果:hMLH1在食管鳞状细胞癌中的阳性表达率(14.92%)显著低于正常食管黏膜组织(95.00%)、非典型增生组织(71.43%)、原位癌组织(68.00%),组间比较差异具有统计学意义(P<0.05)。转化生长因子β受体Ⅱ在食管鳞状细胞癌中的阳性表达率(9.52%)显著低于正常食管黏膜组(85.00%)非典型增生组织(61.90%)、原位癌组织(64.00%)组间比较差异具有统计学意义(P<0.05)。hMLH1 和转化生长因子β受体Ⅱ阳性表达率与肿瘤的分化程度密切相关;且两者在食管癌组织中的表达具有相关性。结论:hMLH1和转化生长因子β受体Ⅱ的异常表达与食管鳞状上皮癌变有关;转化生长因子β受体Ⅱ可能是hMLH1的靶基因。

     

    Abstract: Objective:To investigate the significance and relation of transforming growth factor-β type Ⅱ receptor gene and hMLH1 gene in carcinogenesis of esophageal mucosa epithelium. Methods:The expression of transforming growth factor-β type Ⅱ receptor gene and hMLH1 gene in normal esophageal mucosa epithelium and variant lesions were detected by immunohistochemistry,and their correlation with clinicopathological parameters were analysed as well. Results:The positive expression rate (14.29%) of hMLH1 in esophageal squamous cell carcinoma is significantly lower than that in normal esophageal mucosa epithelium (95.00%),atypical hyperplasia(71.43%), carcinoma in situ (68.00%)(P<0.05);which showed statistical significance. The positive expression rate (9.52%) of TGFβRⅡ in esophageal squamous cell carcinoma is significantly lower than that in normal esophageal mucosa epithelium(85.00%),atypical hyperplasia (61.90%), carcinoma in situ (64.00%)(P<0.05);which showed statistical significance. The positive expression of hMLH1 and Transforming growth factor-β type Ⅱ receptor had close correlation with the differentiation of esophageal carcinoma;the expression of hMLH1 and TGFβRⅡ in esophageal squamous cell carcinoma had correlation. Conclusion :The abnormal expression of hMLH1 protein may be related with the carcinogensis of the esophageal squamous cell. Transforming growth factor-β type Ⅱ receptor may be the target gene of hMLH1.
