人肺腺癌诺维本耐药细胞株的耐药机制

Investigation on Mechanism of Drug Resistance of Human Lung Adenocarcinoma Drug-resistant Cell Line Anip973/NVB

  • 摘要: 研究人肺腺癌诺维本耐药细胞系Anip973/NVB的耐药机制。 方法 免疫组织化学法检测人肺腺癌诺维本耐药细胞系的P-糖蛋白(P-gp)、多药耐药相关蛋白(MRP)的表达,逆转录-聚合酶链反应(RT-PCR)法检测MDR1基因以及MRP3基因的转录。 结果 Anip973/NVB中MRP呈强阳性表达,而Anip973未见表达。Anip973/NVB细胞中MRP3的mRNA转录增加,未检测到MDR1的mRNA产物(P<0.05)。 结论 MRP3在人肺腺癌诺维本耐药细胞中高表达,说明诺维本的耐药机制可能与MRP3的表达相关。

     

    Abstract: Objective To study the mechanism of drug resistance of human lung adenocarcinoma multidrug-resistant cell line Anip973/NVB. Methods Immunohistochemical technique was used to detect the expression of P-gp and MRP.Gene expression of MDR1 and MRP3 were detected by RT-PCR. Results The overexpression of MRP was detected in Anip973/NVB cells,but not in Anip973 cells.The RT-PCR results showed in Anip973/NVB cells the transcription al level of MRP3 was higher than that of control and no transcription of MDR1 was detected. Conclusion MRP3 was overexpressed in lung adenocarcinoma drug resistant cells line Anip9731 NVB.The mechanism of drug resistance of Anip973/NVB may be relatied with the overexpression of MRP3.
