食管鳞状细胞癌中Wnt2、β-catenin、c-myc 和cyclinD1的表达及临床意义

Expression and Significance of Relevant Factors of Wnt2,β-catenin,c-myc and cyclin D1 in Esophageal Squamous Cell Carcinoma Tissue

  • 摘要: 目的研究Wnt2、β-catenin及其下游的靶基因c-myc、cyclinD1在食管鳞癌中的表达情况,以期探讨其在食管鳞癌发生发展中的作用及意义。方法用免疫组织化学SP法检测40例食管鳞癌患者石蜡包埋的癌组织、癌旁组织及正常组织中Wnt2、β-catenin、c-myc、cyclinD1表达情况,分析Wnt2、β-catenin和c-myc、cyclinD1表达之间的相关性及与临床病理特征的关系。结果Wnt2、β-catenin和c-myc、cyclinD1在食管癌组织、癌旁组织及食管正常组织中表达阳性率逐渐降低,差异有统计学意义(P<0.01),β-catenin和c-myc与肿瘤浸润和有无淋巴结转移相关(P<0.05)。结论Wnt2β-catenin Tcf4通路在食管鳞状细胞癌发生发展中起重要作用,该通路有可能成为基因治疗的潜在靶点。

     

    Abstract: To investigate the expression of Wnt2,β-catenin and their target genes such as c-myc,cyclinD1 in tissues of esophageal squamous cell carcinoma (ESCC). Methods The expression level of Wnt2,β-catenin,c-myc, cyclin D1 in 40 cases of esophageal squamous cell carcinoma was detected by immunohistochemistry,and its correlation with clinicopathological features was analyzed. Results The aberrant expressions of Wnt2,β-catenin,c-myc and cyclinD1 in ESCC was higher than tissue adjacent to cancer and normal tissues, there was a significant difference among the groups(P<0.01). The expressions of β-catenin and c-myc protein were closely correlated with infiltrative depth and lymphatic metastasis in esophageal squamous cell carcinoma ESCC(P<0.05). Conclusion Wnt2β-catenin Tcf4 signal pathway plays an important role in ESCC and may become a prognostic factors or gene therapy target for ESCC.
