Effects of MT Overexpression on Drug Resisatnce in Breast Cancer Cells

  • 摘要: 目的 研究金属硫蛋白 ( Metallothionein,MT)基因过表达对乳腺癌细胞耐药性的影响。方法 表达质粒 p BAc Neo MT A转染 MCF- 7/Adr细胞, 用 SRB方法检测了 MT过表达细胞对顺铂的耐药性的变化。结果 与对照细胞相比, p BAc Neo MT A转染的细胞对顺铂的耐受性提高, 其 LD50 由约 1 2 u M增加到大约 30 u M。结论  MT过表达可提高乳腺癌细胞对顺铂的耐药性.

     

    Abstract: Objective To study the effects of MT(Metallothionein)overexpression on drug resistance in breast cancer cells. Methods Expression plasmid pBAcNeoMT ⅡA was transfected into breast cancer cell lineMCF 7/Adr cells.The drug resistance of the cells to cisplatin were detected by SRB experiment. Results Compared to the control cells,MCF 7/Adr cells transfected with MTⅡA cDNA showed enhanced resistance to cisplatin,The LD 50 increases from about 12 uM to approximately 30uM.
