N F- КB 在脂肪酸合酶抑制剂诱导人结肠癌LoVo 细胞凋亡中的作用

Role of N F- КBin Fatty Acid Synthase Inhibitor Induced Apoptosis in Human Colonic Cancer Cells colonic cancer cells

  • 摘要: 目的 探讨 NF-κB在脂肪酸合酶抑制剂 ( FASI)诱导人结肠癌 Lo Vo细胞凋亡中的作用。方法  FASI—浅蓝菌素处理大肠癌 Lo Vo细胞株, 四唑盐比色试验 ( MTT法 )、片断 DNA琼脂糖凝胶电泳、流式细胞仪及荧光染色 ( AO/EB)等方法检测细胞调亡, 用凝胶电泳迁移率法 ( EM-SA)检测 NF- κB活性。结果  Lo Vo细胞在 FASI作用下, 细胞增殖被阻遏, 并呈现剂量效应关系。同时诱导细胞发生凋亡。FASI对人成纤维细胞增殖无明显影响。NF- κB在不同浓度浅蓝菌素 ( 1 0 -9~ 1 0 -5)处理 Lo Vo细胞 1 2 h后, 可抑制 NF- κB活性, 其抑制程度呈剂量依赖性递减。结论FASI可能通过抑制 Lo Vo细胞内源性脂肪酸合成, 诱发细胞凋亡来阻遏 Lo Vo的增殖。NF- κB可能参与 FASI诱导人结肠癌 Lo Vo细胞凋亡中的信号转导.

     

    Abstract: Objective To explore the role of nuclear factor KappaB(NF-κB)in fatty acid synthase inhibitor(FASI),cerulenin,induced apoptosis in human colonic cancer cells.
