人参皂苷Rh2对荷肺癌小鼠基因表达的肿瘤特异性调节

Tumor Specific Gene Regulation by Ginseng Saponin Rh2 in Mice Bearing Lung Cancer

  • 摘要: 目的研究人参皂苷Rh2对正常和荷肺癌小鼠基因表达的调节差异。方法运用寡核苷酸芯片技术,检测Rh2对正常和荷瘤小鼠的基因表达调节,并对差异基因进行聚类和统计分析。为了进一步验证芯片结果,我们使用实时荧光定量PCR方法检验Rh2对基因表达的作用效果。结果与正常小鼠相比,Rh2对荷瘤小鼠的基因表达影响更大,二者的主要受调脏器也不同;Rh2对正常和肿瘤小鼠基因表达调节没有明显的相关性;Rh2能有效拮抗局部接种肿瘤引起的荷瘤小鼠各脏器基因异常表达。结论Rh2对荷瘤小鼠的基因表达调节具有肿瘤特异性。

     

    Abstract: Objective To study gene differential expression profiles by ginsenoside Rh2 in normal and tumor-bearing mice. Methods Gene expression profiles after treatment with Rh2 in normal and tumor-bearing mice were determined by Oligo chip technology as well as the clustering and statistic analysis of single gene.To further validate the microarray results,we used quantitative real-time RT-PCR to examine the effect of Rh2 on individual genes. Results As compared with normal mice,treatment with Rh2 changed gene expression more potently in mice bearing tumor. Thereps no obvious correlation on the gene expression profiles between the normal and tumor2bearing mice. Treatment with Rh2 was found to be effectively resistant abnormal gene expression profiles in some major organs that resulted f rom the local implantation of tumor. Conclusion  The data showed that there is specific regulation of gene expression profiles related to tumor by Rh2 in mice bearing tumor.
