苯丙氨酸解氨酶脂质体的制备及其抗白血病细胞增殖作用的初探

The preparation of liposome of phenylalanine ammonia-lyase and its cytotoxic activity assay on leukemia cells

  • 摘要: 目的 探讨苯丙氨酸解氨酶 (PAL)脂质体对小鼠白血病L12 10 细胞和人白血病K562 细胞生长的抑制作用。方法 采用改良的逆相蒸发法制备出PAL脂质体, 应用MTT还原法分别检测PAL脂质体和游离的PAL对L12 10 和K562 细胞增殖的作用。结果 在酶活性相同条件下, PAL脂质体比游离PAL有较强的抑制细胞增殖作用, 对K562 细胞作用 4 8h, IC50 分别为 0 .90 5U/ml和 1.14 0U/ml,对L12 10 细胞作用 4 8h, IC50 分别为 0 .95 8U/ml和 1.5 13U/ml。结论 PAL脂质体能抑制L12 10 和K562 白血病细胞增殖, 并呈时间和剂量依赖关系 (P <0 .0 1)。

     

    Abstract: Objective The effect of liposomal phenylalanine ammonia-lyase(PAL) on inhibiting the growth of mouse leukemia L_ 1210 cells and human leukemia K_ 562 cells was studied in vitro. Methods The liposome of PAL was prepared by a modified reverse-phase evaporation method. The inhibitory effects of liposomal PAL and free-PAL on leukemia cells were investigated by MTT colorimetric assay separately.
