茶多酚对肝癌细胞生长及端粒酶活性抑制的研究

The Inhibition Effect of Green Tea Polyphenoles on Proliferation and Telomerase Activity of the BEL-7402 Human Hepa tocellular Carcinoma Cell

  • 摘要: 目的 探讨茶多酚在体外对肝癌细胞 BEL- 740 2生长的抑制作用及抗癌机制。方法 MTT法测定茶多酚的对人肝癌细胞生长抑制作用, 集落形成实验观察药物的诱导分化作用, PCR-ELISA试剂盒测定细胞的端粒酶活性。结果 茶多酚能抑制肝癌 BEL- 740 2细胞的增殖, 其 IC50 为1 5 0μg/ml,且具有诱导分化作用, 可降低端粒酶活性的表达, 对 p5 3具有下调作用。结论 茶多酚具有体外抗肿瘤活性, 其抗癌机制可能是通过抑制端粒酶的活性而实现的。

     

    Abstract: Objective To study the anti cancer activity of green tea polyphenoles on BEL 7402 human hepatocellular carcinoma cell and inhibition on telomerase activity in vitro. Methods Cytotoxicity assay was tested by MTT method.Signs of induction differentiation was observed by colony forming units assay.Telomerase activity in cell was determined by PCR ELISA method.
