膀胱肿瘤细胞对T淋巴细胞功能抑制的研究

The Role of Bladder Cancer Cell in T Cell Functional Inhibition in Vitro

  • 摘要: 目的 了解膀胱肿瘤细胞对T淋巴细胞功能的影响。方法 将膀胱肿瘤细胞株EJ、T2 4, 接受致死性射线的EJ、T2 4, 人正常膀胱移行上皮分别与由正常人外周血中分离所得的T淋巴细胞共培养, 2 4小时后分离共培养的T淋巴细胞, 行T淋巴细胞增殖反应 (MTT法 )及T淋巴细胞凋亡检测(TUNEL法 )。结果 EJ组和T2 4组分别与接受致死性射线的EJ组和T2 4组相比, T淋巴细胞的增殖反应降低 (P <0 .0 1) ;TUNEL法检测T淋巴细胞凋亡结果的比较 :EJ组和T2 4组分别与接受致死性射线的EJ组和T2 4组比较, T淋巴细胞凋亡指数均降低 (P <0 .0 5 )。结论 膀胱肿瘤细胞株EJ、T2 4可通过某种机制抑制T淋巴细胞的功能, 增加T淋巴细胞凋亡。

     

    Abstract: Objective To research the role of bladder cancer cell in T cell functional inhibition in vitro. Methods Co-culture EJ,T24(the bladder cancer cell line),lethal irradiated EJ, lethal irradiated T24, bladder epithelial cell with T cell individually for twenty four hours, isolating T cell, measuring the function and apoptosis index of T cell with MTT and TUNEL method.
