钙拮抗剂逆转L1210/HR细胞抗药性的动物体内试验研究

Study on Reversal of Resistance to Homoharringtonnine (HH) by Calcium Antagonist in DBA/2 Mouse Transplantable L1210 Leukemia Cells

  • 摘要: 本文将体外培养的抗高三尖杉酯碱L1210耐药细胞(L1210/HR细胞)移植至DBA/2小鼠体内,建立了L1210/HRDBA/2小鼠动物模型,并用钙拮抗剂异搏定、尼群地平进行L1210/HRDBA/2动物模型的逆转试验,证明异搏定能明显延长动物的存活期,有效地逆转了L1210/HR细胞的抗药性。另外用高效液相色谱议检测了经异搏定、尼群地平逆转的L1210/HR细胞内高三尖杉酯碱的药物含量,表明经钙拮抗剂异搏定逆转后细胞内高三尖杉酯碱药物含量明显增高,经异搏定逆转的细胞内药物含量比未逆转的细胞高2.3倍。

     

    Abstract: HH-resistant DBA/2 mouse L1210(L1210) leukemia modal was established in our laboratory. Calcium antagonists were used to reverse HH resistance of these sublines. The results showed that the servival of mice was prolonged and HH resistance of L1210/HR cells was reversed by verapamil. The concentrations of verapamil and nitrendipine in L1210/HR cells were also measured. After L1210/HR cells reversed by verapamil, the amount of HH was markedly accumulated in L1210/HR sublines.
