超氧阴离子自由基加强肿瘤坏死因子抗膀胱肿瘤作用
Synergistic Activity of TNF-a with Superoxide Anion,SOD or Hydroxyl Radical on Human bladder Cancer

摘要: 目的为阐明活性氧与肿瘤坏死因子α(TNFα)抗肿瘤作用的关系;方法将膀胱癌细胞系BIU-87作体外培养,并将TNF-α和不同浓度的超氧阴离子(O2-)、径自由基(·OH)或超氧化物歧化酶(SOD)作用于细胞,用MTT法测定细胞毒效果;结果显示TNF-α对BIU-87有明显的浓度依赖性抑制作用,O2-或TNF-α单独均对细胞有抑制作用,O2-又可明显加强TNF-α对BIU-87细胞的抑制,SOD(300U/ml)与TNF-α(10000U/ml)作用于细胞后其增殖明显高于单用TNF-α者,进一步证明O2-的加强作用,且说明SOD可保护BIU-87细胞免受TNF-α的抑制,用·OH后的影响则无统计意义;结论超氧阴离子参与及影响TNF-α对膀胱肿瘤的作用,该研究将对探讨TNF-α抗膀胱肿瘤作用的机理提供一定的依据。Abstract: To investigate synergistic effects of tumour necrosis factor-a(TNF-a) with superoxide anion,SOD or hydroxyl radical on human bladder cancer. Human bladder cancer cell line BIU一87 was cultured.The synergistic action of superoxide anion,SOD or hydroxyl radical in combination was tested by MTT assay.The growth of BIU一87 cell was inhibited by TNFa in dose dependent manner.The cytotoxocity of TNF-a to BIU87 cell was enhanced by superoxide anion and inhibited by SOD.However,the combination of TNF-a and hydroxyl radical did not showed this sygnergistic action.This study demostrated that superoxide anion-TNF-a showed synergistic action against the BIU-87 cell line.
