Induction of apoptosis in human Panereatic cancer cell line SW-1990 by hydroxyl camptothecinum

  • 摘要: 目的 :研究羟基喜树碱体外诱人胰腺癌细胞株SW-1990凋亡的作用. 方法 :以不同浓度的羟基喜树碱处理在体外培养的胰腺细胞,以电子显微镜、流式细胞仪和原位末端标记分析和检测细胞凋亡. 结果 :在0.05mg/ml浓度下,羟基喜树碱作用于体外培养的胰腺癌细胞20小时后,肿瘤细胞的凋亡率在13.2%- 15.3%之间,主要以早、中期的表现为主:在0.lmg/ml浓度下,作用20小时后,凋亡率在26.l%-30.4%之间,凋亡形态学表现主要以晚期为主. 结论 :羟基喜树碱可作为细胞凋亡诱导剂用于胰腺癌治疗.

     

    Abstract: Objective To study the effect of hydroxyl camptothecinum on human pancreatic cancer cell apoptosis in vitro. Methods :SW-1990 ce-ll line were treated with hydroxyl camptothecinum at different concentrations.cell apoptosis was evaluated by electric microscopy,flow cytom-etry and in situ TUNEL(TdT-mediated dUTP-X nick end labeling). Results The apoptosis rate of Pancreatic cell treatet with hydroxyl cam-ptothecintm at the concentration of 0.05mg/ml for 20 hours is between 13.2%-15.3%,mainly at the early and middle morphological stage.At the concentration of 0.lmg/ml,the rate is between 26.1%-30.4%,mainly at late stage. Conclusion hydroxyl camptothecinum is a potential agent for pancreatic cancer chemotherpy.
