Apoptosis in HeLa Cells Induced by Antisense Oligodeoxynucleotide of Human survivin Gene

  • 摘要: 目的 探讨survivin 反义核酸诱导HeLa 细胞发生凋亡的可能性,揭示该凋亡发生与Bcl22 和Bax 之间的关系。方法 采用TUNEL 染色法,研究survivin 反义核酸与HeLa 细胞凋亡的关系;通过免疫组织化学法检测凋亡相关基因Bcl22 和Bax 的表达。结果 survivin 反义核酸在体外能诱导HeLa 细胞凋亡,下调Bcl22 的表达,增强Bax 的表达。结论 诱导HeLa 细胞凋亡是survivin 反义核酸抗HeLa 作用的机制之一,survivin 反义核酸可能通过下调Bcl22 的表达,增强Bax 的表达,诱导HeLa 细胞发生凋亡。

     

    Abstract: Objective  To investigate the apoptosis in HeLa cell induced by antisense oligodeoxynucleotide of human survivin gene, and the relation between this apoptosis and expression of Bcl22 and Bax. Methods  In vi t ro experiment s, TUNEL staining method was used to detect the apoptosis status of cervical canc2 er HeLa cell line t reated by antisense oligodeoxynucleotide of survivin. Immunohistochemical staining was used to detect the expression of apoptosis2relation gene Bcl22 and Bax. Results  Antisense oligodeoxy nu2 cleotide of survivin was able to induce the apoptosis in HeLa cells. Antisense oligodeoxynucleotide of sur2 vivin can reduce the expression of apoptosis2relation gene Bcl22 and increase the expression of apoptosis2 relation gene Bax. Conclusion  Antisense oligodeoxynucleotide of survivin was able to induce the apopto2 sis in HeLa cell . This apoptosis may be mediated by down2regulation of apoptosis2regulated gene Bcl 22 and up 2regulation of Bax.
