葡聚糖磁流体热处理对小鼠H22移植瘤生长的影响及机制初探

Primary Study on the Effect of Hyperthermia Treated with Dextran Magnetic Fluid on Grafted H22 Tumor in Mice

  • 摘要: 目的探讨磁流体热处理对小鼠H22移植瘤生长及血管内皮生长因子(VEGF)表达和凋亡状况的影响。方法将接种了H22肝癌细胞的小鼠随机分为4组。对第4组移植瘤内注射葡聚糖磁流体,在55kHz,20kA/m交变磁场中作用10min。第1、第2、第3组作为对照,分别对瘤内注射盐水不加磁场,注射盐水加磁场和注射磁流体不加磁场。两周后处死动物,称瘤重,计算抑瘤率。另两组动物分别瘤内注射磁流体和等体积盐水,作为处理组和对照组,在经磁场处理后1h、5h、24h、48h和336h处死,检测瘤组织的VEGF表达和凋亡状况。结果第4组移植瘤内的温度可达到46℃~48℃。与第1、第2、第3组相比,第4组抑瘤率分别为:59.74%、52.51%和40.35%,其中与第1组差异有显著性(P<0.05)。磁热处理后不同时间,处理组肿瘤组织的VEGF表达和凋亡状况与对照组的比较差异均无显著性。结论单次磁流体热处理可以显著抑制小鼠H22肝癌移植瘤的生长。抑制VEGF表达和诱导凋亡不是抑制移植瘤生长的主要原因。
