宫颈癌组织高危HPV18 L1基因的克隆及序列分析

Cloning and Sequencing of Human Papillomavirus Type 18 L1 Gene from Cervical Cancer Tissue

摘要: 目的克隆和分析人乳头瘤病毒18型晚期表达基因L1序列,为HPV感染的检测和基因工程疫苗研制提供基础。方法采用PCR技术扩增了1例HPV18阳性宫颈腺癌患者癌组织中HPV18L1基因,并对其进行了克隆及全序列分析。结果本例宫颈癌临床标本HPV18L1基因与GenBank收录的原型比较有12个碱基变异,推导其编码的氨基酸也发生了变化。结论本例中国人宫颈癌HPV18L1基因有一定的变异。

Abstract: Objective To clone and analyze the whole sequence of L1 gene and its deduced L1 protein of HPV18 from specimen of cervical cancer. Methods HPV18 L1 gene was amplified from a HPV18-positive DNA sample of a Chinese patient with cervical adenocarcinoma by means of PCR and then cloned and sequenced.
