三七总甙对NB4细胞核转录因子-κB及组织因子表达的影响

摘要: 目的 通过核转录因子NFκB及其抑制因子IκB的表达探讨三七总甙 (PanaxnotoginsengsaponinsPNS)对NB4细胞组织因子的调控机制。方法 用三七总甙处理NB4细胞, 分别于处理后 2 4h、4 8h、72h、96h、12 0h收获细胞, 半定量逆转录PCR检测TF mRNA的表达 ;Western蛋白印迹检测细胞内NFκB的亚单位 p6 5及IκBα的蛋白, 以及核蛋白p6 5的表达。 结果 三七总甙处理NB4细胞 2 4h, TF mRNA表达开始下降, 12 0h完全抑制。Westernblot显示三七总甙抑制细胞内IκBα及核蛋白 p6 5的表达。结论 三七总甙抑制组织因子的表达, 可能是通过抑制NFκB的活化起作用。Abstract: Objective To investigate the effects of Panax notoginseng saponins(PNS)on nuclear factor κB and tissue factor in NB4 cells. Methods The effects of PNS on TFmRNA,p65 and IκBα expression were evaluated by RT-PCR and Westernblotting,respectively. Results PNS down-regulated the expression of TF-mRNA. PNS inhibited the expression of IκBα and nuclear p65. Conclusion PNS could down-regulate the expression of TF-mRNA by inhibition of NFκB pathway.
