Abstract:
Objective To investigate the expression levels of BRD7 and NGX6 gene in bone marrow cells from adult patients with acute leukemia(AL). Methods The bone marrow mononuclear cells (BMNC) obtained from 52 patients with acute leukemia(AL) and 30 normal bone marrow subjects as controls were separated by density gradient centrifugation. The mRNA levels of BRD7/NGX6 in the BMNCs were detected by RT-PCR. The expression of BRD7 protein in 7 patients with AL was assayed by immunohistiochemical stains. Results The mRNA expression of BRD7/ NGX6 in BMNCs f rom patients with AL and cont rols were detected. The mRNA relative expression of N GX6 gene was no significant difference in both AL and the cont rol group (

P = 0. 982) . The mRNA expression of BRD7 in patients with AL was higher than that in the cont rol group, and the difference was significant (

P = 0. 001), but there was no significant difference in ALL, M1, M2, M3, M4 and M5 subtypes of ANLL (

The mRNA expressions of BRD7 gene in patient s with AL at onset were not correlated with blast cells rates in bone marrow cells, and the same as platelets and white blood cells count s in total peripheral blood. The protein expression of BRD7 was detected in part samples, and there were high percentage of BRD7 protein positive cells in patient s with AL. Conclusion The expression of BRD7/ N GX6 were detected in BMNCs f rom patient s withAL and normal subjects, and the expression of BRD7 was up-regulated in AL cells. But the mRNA expression level of BRD7 gene was not correlated with some clinical character of patient s with AL at onset .
