Abstract:
Objective To study the influence of gefitinib, a selective epidermal growth factor receptor tyrosine kinase inhibitor, on the radiosensitivity of human cervical carcinoma cells in vitro. Methods The effect of gefitinib on cervical carcinoma cells ( Siha, HeLa) was determined by MTT assay. Cells were assayed for clonogenic survival to determine the radiosensitization effect induced by gefitinib. Results The proliferation of cervical carcinoma cell lines (Siha, HeLa) was inhibited by gefitinib, and the effect was dose-dependent . The value of IC
20 for the effect of gefitinib on cervical carcinoma cell lines (Siha, HeLa) was 0. 15μmol/ L and 0. 32μmol/ L respectively. When exposed to gefitinib for 72h, the SER of Siha and HeLa cell was 1. 693 and 1. 228 respectively. Conclusion The selective epidermal growth factor receptor tyrosine kinase inhibitor gefitinib can induce radiosensitization of cervical carcinoma cells.
