Research Progress and Challenges in CAR-T Cell Therapy for Solid Tumors

  • 摘要:   嵌合抗原受体 T 细胞(Chimeric Antigen Receptor T-Cell,CAR-T)疗法作为肿瘤免疫治疗领域的革命性技术,已在血液系统恶性肿瘤治疗中取得突破性进展,目前全球已有多款 CAR-T 产品获批临床应用。然而,该疗法在实体瘤治疗中的进展相对缓慢,主要归因于实体瘤具有高度复杂的生物学特征。近年来,随着实体瘤新靶点的不断挖掘、基因工程技术的持续创新以及联合治疗策略的日益成熟,CAR-T 疗法在实体瘤治疗中的应用潜力逐步显现,展现出广阔的转化前景。本文系统综述实体瘤 CAR-T 疗法的最新临床研究进展,深入分析其面临的核心挑战及应对策略,并对该领域的未来发展方向进行展望。

     

    Abstract:   
      Chimeric Antigen Receptor T-cell (CAR-T) therapy, a revolutionary technology in tumor immunotherapy, has achieved breakthroughs in the treatment of hematological malignancies. Currently, multiple CAR-T products have been approved for clinical use worldwide. However, its progress in solid tumor treatment is relatively slow, mainly due to the highly complex biological characteristics of solid tumors. In recent years, with the continuous identification of new targets in solid tumors, constant innovation of genetic engineering technologies, and increasing maturity of combination therapy strategies, the application potential of CAR-T therapy in solid tumor treatment has gradually emerged, showing broad translational prospects. This article systematically reviews the latest clinical research progress of CAR-T therapy for solid tumors, deeply analyzes the core challenges it faces and corresponding countermeasures, and provides an outlook on the future development direction of this field.
