Expression and Clinical Significance of miRNA-152 in Hepatocellular Carcinoma

    目的 探讨微小RNA(miR)-152在肝癌组织中的表达及临床意义。
    方法 收集56例肝癌患者癌及癌旁组织标本,采用实时荧光定量聚合酶链反应技术(Real-time PCR)检测肝癌及癌旁组织标本中miR-152的表达水平,分析其与肝癌患者临床病理特征及预后的相关性。
    结果 miR-152在肝癌及癌旁组织中相对表达水平分别为0.616±0.041及0.768±0.042,两者差异有统计学意义(P=0.011);其与患者的TNM分期(P=0.042)、病理分化(P=0.035)、肿瘤转移(P=0.048)及血管侵犯(P=0.042)明显相关,而与性别、年龄、AFP水平、肿瘤个数、肿瘤直径等无明显相关;miR-152低表达组患者总生存时间(P=0.035)和无进展生存时间(P=0.012)明显低于miR-152高表达组,多因素Cox回归分析的结果显示,miR-152低表达、肿瘤复发和病理分化是影响肝癌患者预后的独立危险因素。
    结论 miR-152在肝癌组织中表达下调,且与肝癌患者的病理分化程度、TNM分期、有无转移、有无血管侵犯及预后密切相关。

     

    Abstract:
    Objective To investigate the expression of microRNA-152 in hepatocellular carcinoma, and analyze the correlation of miR-152 expression with the clinicopathological characteristics of hepatocellular carcinoma(HCC) patients.
    Methods Real-time PCR was used to detect the expression of miR-152 in tissue specimens from HCC and the adjacent non-cancerous hepatic tissues collected from 56 patients who underwent surgical treatment for primary HCC. The association between the expression of miR-152 and the clinical pathological characteristics and prognosis of HCC patients was analyzed.
    Results The relative expression of miR-152 in hepatocellular carcinoma and corresponding adjacent non-carcinoma tissues were 0.616±0.041 and 0.768±0.042 (P=0.011). The expression of miR-152 was significantly associated with TNM stages (P=0.042), tumor differentiation (P=0.035), metastasis (P=0.048) and venous invasion (P=0.042). There was no significant association between the miR-152 expression and gender, age, AFP level, tumor number or tumor grade. The OS (P=0.035) and PFS (P=0.012) in the low miR-152 expression group were obviously shorter than those in the high miR-152 expression group. Multivariate Cox regression analysis showed that the low expression of miR-152, tumor recurrence and pathological classification were significant independent prognostic factors for the prognosis of hepatocellular carcinoma patients.
    Conclusion The expression of miR-152 is decreased in hepatocellular carcinoma tissues, and the low expression of miR-152 is significantly associated with TNM stages, tumor differentiation, metastasis, venous invasion and the prognosis of HCC patients.
