Expression and Methylation of PDCD4 in Gastric Carcinoma and Its Clinical Significance

    目的 观察胃癌组织中抑癌基因PDCD4启动子区甲基化状态及其对PDCD4表达水平的影响并探讨其临床意义。
    方法 通过免疫组织化学、Western blot法检测胃癌组织中PDCD4蛋白的表达;RT-PCR法检测PDCD4 mRNA的表达;甲基化特异性PCR(MSP)法检测PDCD4启动子区甲基化水平。分析PDCD4表达水平以及启动子甲基化水平与胃癌患者临床病理特征之间的相关性。
    结果 胃癌组织中PDCD4蛋白及mRNA表达水平均显著降低(P < 0.05),甲基化作用显著增强(P < 0.05)。PDCD4蛋白表达缺失与胃癌的分化、临床分期以及淋巴结转移密切相关(P < 0.05);PDCD4高甲基化与胃癌的淋巴结转移、临床分期密切相关(P < 0.05)。PDCD4甲基化水平与PDCD4蛋白以及mRNA表达水平均呈负相关(P < 0.05)。
    结论 胃癌组织中PDCD4表达水平显著降低,并与胃癌发展相关,启动子区高甲基化可能是PDCD4表达缺失的原因。

     

    Abstract:
    Objective To observe the methylation status of the promoter region of the tumor suppressor gene PDCD4 in gastric cancer tissues and its effect on the expression level of PDCD4, and to explore its clinical significance.
    Methods The protein expression of PDCD4 in gastric cancer tissues was detected by immunohistochemistry (IHC) and Western blot; the mRNA expression of PDCD4 was detected by RT-PCR; the methylation level of PDCD4 promoter region was detected by MSP. The correlation between PDCD4 expression, promoter methylation level and clinicopathological characteristics of gastric cancer patients was analyzed.
    Results The expression levels of PDCD4 protein and mRNA in gastric cancer tissues were significantly decreased (P < 0.05), and the methylation of PDCD4 was significantly enhanced (P < 0.05). Deficiency of PDCD4 protein expression is closely correlated with the differentiation, clinical stage and lymph node metastasis of gastric cancer (P < 0.05); hypermethylation of PDCD4 is closely correlated with lymph node metastasis and clinical stage of gastric cancer (P < 0.05). The methylation level of PDCD4 was negatively correlated with the protein and mRNA expression levels of PDCD4 (P < 0.05).
    Conclusion The expression of PDCD4 in gastric cancer tissues is significantly decreased and correlated with the development of gastric cancer. Hypermethylation of CpG island in promoter region may be the reason for the deficiency of PDCD4 expression.
