Expression and Correlation of miR-135a-5p, GATA3 and STAT3 in Cervical Cancer

    目的 探讨不同病理类型宫颈组织及宫颈癌细胞系中miR-135a-5p、GATA3和STAT3的表达及其与宫颈癌之间的关系。
    方法 实时定量RT-PCR法检测各宫颈组织及细胞中miR-135a-5p的表达水平;免疫组织化学SP和Western blot法检测宫颈组织和细胞中GATA3和STAT3蛋白的表达;双荧光素酶报告基因验证miR-135a-5p与GATA3 3’UTR间的作用,分析miR-135a-5p和GATA3 mRNA的表达结果与宫颈癌临床病理参数的关系。
    结果 miR-135a-5p在宫颈癌组织及细胞系中的表达均上调(均P < 0.05)。GATA3 mRNA的表达水平随着宫颈病变程度增加而降低(P < 0.05);STAT3 mRNA表达水平随着宫颈病变程度增加而升高(P < 0.05)。miR-135a-5p可靶向作用于GATA3 3’UTR。宫颈癌组织中miR-135a-5p与GATA3(r=-0.6656)、GATA3和STAT3 mRNA表达水平呈负相关(r=-0.4534)。在宫颈癌中miR-135a-5p表达水平与临床分期、淋巴结转移有关(P < 0.05);GATA3 mRNA的表达与临床分期、肌层浸润深度及淋巴结转移均有关(P < 0.05)。
    结论 宫颈癌中miR-135a-5p和STAT3表达上调,GATA3表达下调,三者可能通过相互作用参与宫颈癌的发生、发展。

     

    Abstract:
    Objective To investigate the expressions of miR-135a-5p, GATA3 and STAT3 in different pathological types of cervical tissues and cervical cancer cell lines and their correlation in cervical cancer.
    Methods Real-time PCR was performed to determine the expression of miR-135a-5p in cervical carcinoma tissue and cell lines. Immunohistochemistry SP and Western blot methods were used to determine the expression of GATA3 and STAT3 in cervical tissues and cells. The effect of miR-135a-5p on GATA3 3'UTR was verified by dual-luciferase reporter assay system. The correlation of miR-135a-5p and GATA3 mRNA expression with the clinical pathological parameters of cervical squamous cell carcinoma patients was analyzed.
    Results The miR-135a-5p expression was up-regulated in CSCC tissues and cell lines(all P < 0.05). With the increasing stage of carcinoma, the mRNA levels of GATA3 was reduced(P < 0.05), whereas the mRNA levels of STAT3 was increased(P < 0.05). miR-135a-5p could target GATA3 3'UTR. The expression of GATA3 mRNA was negatively correlated with STAT3 mRNA expression(r=-0.4534) and miR-135a-5p expression(r=-0.6656). Moreover, the miR-135a-5p expression was related to clinical stages and lymph node metastasis(P < 0.05), and the GATA3 mRNA expression was related to clinical stage, depth of invasion and lymph node metastasis in cervical squamous cell carcinoma(P < 0.05).
    Conclusion In cervical cancer, the expression of miR-135a-5p and STAT3 mRNA are up-regulated, the expression of GATA3 mRNA is down-regulated, suggesting that they may be involved in the occurrence and development of cervical cancer through interaction.
