Abstract:
Objective To investigate the effect of long-chain non-coding RNA FILNC1 targeting c-Myc expression on the biological behavior of renal cell carcinoma.
Methods qPCR was used to detect the expression of FILNC1 in renal cell carcinoma tissues and different renal cell carcinoma cell lines. Double luciferase reporter gene was used to detect the interaction between FILNC1 and c-Myc. Western blot was used to detect the effect of FILNC1 on c-Myc protein. MTT cell proliferation assay was used to detect the effect of FILNC1 and c-Myc on the proliferation of renal cell carcinoma cells. The effect of FILNC1 and c-Myc on the apoptosis of renal cell carcinoma cells was detected by flow cytometry. Effect of FILNC1 on xenograft differentiation of renal cell carcinoma in nude mice was detected.
Results Compared with other renal cell carcinoma cell lines, the expression level of FILNC1 in A498 cells was significantly higher. Double luciferase assay confirmed that FILNC1 could directly regulate the expression of c-Myc and fluorescence activity. The inhibition of FILNC1 and c-Myc expression could reduce the proliferation of renal cell carcinoma cells and promote cell apoptosis. Inhibition of FILNC1 expression in renal cell carcinoma cells in nude mice in vivo resulted in a certain inhibition of xenograft capacity.
Conclusion FILNC1 could target the expression of c-Myc to influence the proliferation and apoptosis of renal cell carcinoma cells.
