摘要: 目的 分析细胞骨架相关蛋白SM22α在正常胃黏膜及胃癌组织中的表达水平。方法 应用RTPCR 及Western blot印迹检测胃癌及正常胃黏膜组织中SM22α的mRNA和蛋白表达水平。RT-PCR和明胶酶图检测胃癌及正常胃黏膜组织中MMP9的mRNA水平和活性。结果 胃癌组织中SM22α表达水平低于癌旁正常胃黏膜组织(P<0.05),SM22α低表达的胃癌组织中MMP9 mRNA水平和酶活性显著高于对照组(P<0.05),胃癌组织中SM22α表达水平与MMP9活性呈负相关。结论 SM22α在胃癌组织中表达下调,并可能通过负性调节MMP9表达抑制胃癌转移。Abstract: Objective To investigate the expression levels of cytoskeleton-related protein SM22α in gastric cancer or adjacent normal gastric mucosa tissues. Methods RT-PCR and Western blotting were performed to detect SM22α mRNA and protein levels in gastric cancer tissues and adjacent normal gastric mucosa tissues. RT-PCR and zymography were used to detect MMP9 mRNA level and activity in gastric cancer tissues and adjacent normal gastric mucosa tissues, respectively. Results Both SM22α mRNA and protein levels in gastric cancer were significantly lower than that in adjacent normal gastric mucosa tissues (P<0.05). The expression and activity of MMP9 in gastric cancer cases were signifi cant higher than that in adjacent normal gastric mucosa tissues (P<0.05). A negative correlation was found between SM22α protein level and MMP9 activity in gastric cancer tissues. Conclusion SM22α is downregulated in gastric cancer. SM22α may probably inhibit metastasis through negative regulation of MMP9 expression in gastric cancer.
