Abstract:Objective To investigate the expression of microRNA-375 (miR-375) and 3-phosphoinositide-dependent protein kinase-1 (PDK1) in pancreatic cancer and analyze their correlation. Methods miR-375 and PDK1 expression in pancreatic tissues and cell lines were evaluated using quantitative real-time PCR (qRT-PCR).After transfecting miR-375 mimics into Panc-1 cells,the expression of PDK1 mRNA and protein were assessed by qRT-PCR and Western blotting,respectively. Results Significant downregulation of miR-375 in pancreatic cancer and Panc-1 cells was identified,whereas PDK1 was upregulated in these samples.Transfection of miR-375 mimics led to downregulation of PDK1 in Panc-1 cells. Conclusion miR-375 may be a tumor suppressor in pancreatic cancer and regulate PDK1 negatively.
