Expression and Significance of TRX-2 in Bone Marrow of Acute Leukemia

  • 摘要: 目的 探讨TRX-2基因表达水平与急性白血病及临床特征的关系。方法 应用适时定量聚合酶链反应(Real-time quantitative polymerase chain reaction,RT-PCR)和间接免疫荧光试验检测试验组120例急性白血病和对照组120例非血液病的其他疾病患者骨髓TRX-2基因表达情况,并结合临床资料进行分析。结果经RT-PCR检测,试验组TRX-2 mRNA骨髓表达量明显高于对照组,两组差异具有统计学意义[(5.08±0.52)×102 vs.(0.91±0.15)×102,P<0.05]。其中试验组中原始细胞≥90%的样本TRX-2表达量比原始细胞<90%患者明显增多,其差异具有统计学意义[(6.79±0.51)×102 vs.(3.26±0.58)×102,P<0.05]。而骨髓中TRX-2的表达量与年龄、性别和急性白血病的类型无相关性;间接免疫荧光试验检测试验组骨髓原始细胞不同程度表达TRX-2,阳性表达率78.2%,而对照组无阳性表达;TRX-2的RT-PCR检测与间接免疫荧光法检测两种方法之间具有显著相关性(P<0.05),但一致性较差(κ=0.26)。结论TRX-2 在急性白血病骨髓中呈现高表达,其与急性白血病原始细胞的负荷量存在相关性,而与临床特征无明显相关;RT-PCR与间接免疫荧光法两种方法检测TRX-2之间具有显著相关性,但一致性较差。

     

    Abstract: Objective To investigate the correlation between TRX-2 expressions level and clinicopathological charateristics of acute leukemia. Methods The expressions level of TRX-2 in bone marrow from 120 patients with acute leukemia were detected by RT-PCR(Real-time PCR) and indirect immunofluorescence,respectively,and followed by subsequent clinical characteristics correlative studies. Results By RT-PCR detecting,the expressions level of TRX-2 in 120 cases of acute leukemia were significantly higher than that of 120 other cases of free-blood disease[(5.08±0.52)×102 vs.(0.91±0.15)×102, P<0.05].With respect to the percentage of blast cell in bone marrow,an increase trend of TRX-2 mRNA expressions in blast cell of bone marrow over 90% was found in comparison to patients with blast cell of bone marrow bellow 90%,and statistical difference was found between them [(6.79±0.51)×102 vs.(3.26±0.58)×102,P<0.05].TRX-2 mRNA expressions level in bone marrow were not related to age,gender and classifications of acute leukemia.By indirect immunofluorescence detecting,TRX-2 protein in test group occure more frequently than that in control group.There was significant correlation between RT-PCR and indirect immunofluorescence (P<0.05),although with poor consistency(κ=0.26). Conclusion TRX-2 is high expression in acute leukemia and involved in the percentage of blast cell in bone marrow.The presence of TRX-2 appears not related to clinical characteristics.There is significant correlation between RT-PCR and indirect immunofluorescence,although they are poor consistency.
