Effects of miR-199a-3p on Cell Migration and Invasion in Prostate Cancer Cells

  • 摘要: 目的从miR-199a-3p在前列腺癌高、低转移潜能细胞中的表达差异出发,研究其在高转移前列腺癌细胞株1E8细胞运动转移中的作用及可能的分子机制。方法运用Realtime PCR法检测 miR-199a-3p在前列腺癌高、低转移潜能配对细胞系中的表达差异。通过划痕实验及transwell实验观察1E8细胞及转染 miR-199a -3p mimics及mimics NC后该细胞运动迁移能力的变化。结果(1)Realtime PCR结果显示高转移潜能1E8细胞中miR-199a-3p表达水平明显低于低转移潜能2B4细胞。(2)上调miR-199a-3p会减弱1E8细胞的运动转移能力。结论miR-199a-3p的上调可以抑制前列腺癌细胞的运动迁移能力。

     

    Abstract: ObjectiveTo investigate the different expression of miR-199a-3p in different prostate cancer cell lines and the effects of miR-199a-3p on cell migration and invasion in prostate cancer. MethodsThe expression of miR-199a-3p in 1E8 (high metastasis prostate cancer) and 2B4 (low metastasis prostate cancer) cell lines were identified by realtime PCR. The migration and invasion abilities were detected by Transwell experiment.Results(1)By comparing with 2B4 cells, of miR-199a-3p was decreased in 1E8 cells. (2)The invasiveness of 1E8 cells were decreased through miR-199a-3p upregulation. ConclusionmiR-199a-3p invasiveness of prostate cancer cells.
