Abstract:
Objective To study the expression and clinical significance of GKLF and IKLF in 96 human gastric carcinoma tissues and cell lines (MKN-28,SGC-7901,BGC-823,HGC-27).Methods The mRNA levels of GKLF and IKLF in gastric carcinoma tissues and cell lines were determined by real-time fluorescence quantitative PCR.The protein levels of GKLF and IKLF in gastric carcinoma tissues and cell lines were detected by immunohistochemistry and Western blot,respectively.Results The GKLF mRNA level in gastric carcinoma tissues was lower than that in normal gastric mucosa while IKLF mRNA level was higher in the same condition(
P<0.0001).Furthermore,the mRNA levels of GKLF and IKLF in gastric carcinoma tissues were uncorrelated with differentiation.Positive expression rate of GKLF protein in the carcinoma tissues was significantly lower than that in normal gastric mucosa while positive expression rate of IKLF protein was significantly higher(
P=0.001).The expressions of GKLF and IKLF protein were uncorrelated with age,gender,differentiation and Lauren's classification but correlated with lymph node involvement and clinical stage(
P=0.001).The expressions of GKLF mRNA and protein in gastric carcinoma cell lines were lower than that in GES-1 while the expressions of IKLF mRNA and protein were higher(
P<0.05).Conclusion GKLF and IKLF may play roles in the carcinogenesis and progression of gastric carcinoma.To examine the expression of GKLF and IKLF may be useful in diagnose the degree and prognosis of gastric carcinoma.
