Abstract:
Objective To explore the mechanisms of inhibition of artemether (Artm) on glioma growth in brain tumor bearing SD rat. Methods MTT (Methy thiazolyl tetrazolium) assay was used to evaluate the inhibitory effect of artemether treatment on C
6 glioma cells. Forty-eight (24 male and 24 female) SD rats which were subcutaneous planted with SD rat C
6 glioma cell to establish SD rat orthotopic glioma model were divided resourcefully into 6 groups. They were control group; model groups, artemether 33.3 mg/(kg·d), 50 mg/(kg·d), 66.6 mg/(kg·d); two drugs combined group, artm 50 mg/(kg·d)+ ferrous sulfate 1.5 mg/(kg·d) and positive control group. Lomustine 20 mg/(kg·d) SD rats of each group were fed for 10 consecutive days. Inhibitive rates of Artemether were calculated by a
2bπ∕6. Results Artemether could inhibit the proliferation of C
6 glioma cells in concentration and time -depended manners. Inhibitive rates of artemether in different dosages were 54.5%、61.0%、64.5% and 69.8% respectively. Compared among model groups, inhibitive rates of artemether 66.6 mg/(kg·d) was higher than that of artemether 33.3 mg/(kg·d), there was a significant difference between two group (P<0.05). Compared among experiment groups, inhibitive rate of Artm + ferrous sulfate was higher than that of 33.3 mg/(kg·d), there was a significant difference (P<0.05). Conclusion Artemether can inhibit glioma growth in SD rat orthotopic glioma model.
