低氧对白血病细胞株Raji细胞侵袭转移能力的影响

  • 摘要: 目的 研究低氧对白血病细胞株Raji细胞侵袭转移能力的影响。 方法 Raji细胞体外常规培养,用体积分数为1%、3%、5%的氧分别处理Raji细胞24h,以常氧培养Raji细胞为对照组,通过细胞黏附实验检测细胞黏附力,细胞迁移实验检测细胞运动力,肿瘤细胞体外侵袭实验检测细胞侵袭力,RT-PCR检测细胞VEGF、MMP-2、MMP-9 mRNA的表达,Western Blot检测细胞HIF-1α蛋白的表达。 结果 与对照组相比,3%、5%的氧均能增强Raji细胞黏附力、运动力和侵袭力(P<0.05或P<0.01),而且还能上调Raji细胞HIF-1α蛋白、HIF-1α mRNA、VEGF mRNA、MMP-9 mRNA的表达(P<0.01),各组MMP-2 mRNA均无明显表达;而1%氧与对照组相比各检测指标无明显变化(P>0.05)。 结论 适度低氧可增强白血病细胞株Raji细胞侵袭转移能力,其机制可能是通过HIF-1α上调VEGF、MMP-9表达实现的。

     

    Abstract: Objective To study the potential effect of hypoxia on invasion and metastasis of leukemia cell line Raji. Methods Raji cells were cultured with normal method in vitro and treated with 1%,3%,5% oxygen for 24h, the normoxic cultured group was used as control. Then cell adhesion assay,cell migration assay and cell invasion assay were used to observe the effects of oxygen on adhesion, migration and invasion ability of Raji cells, and RT-PCR was used to evaluate the mRNA expression levels of HIF-1α, VEGF, MMP-2 and MMP-9. The protein level of HIF-1α was observed by Western Blot. Results Compared with control group, 3%,5% oxygen groups could significantly enhanced the adhesion,migration and invasion of Raji cells (P<0.01 or P<0.05),and could also up-regulate the protein level of HIF-1α and the mRNA level of HIF-1α,VEGF and MMP-9,but the mRNA level of MMP-2 was not obviously expression. However, all indexes of 1% oxygen group merely changed when compared with control group (P>0.05). Conclusion Moderate hypoxia could enhance the ability of invasion and metastasis of Raji cells. The possible mechanism is HIF-1α up-regulate the expression levels of VEGF and MMP-9.
