Abstract:
Objective To investigate the enhanced anti-tumor effect of the dendritic cells by lycium bararum polysaccharides. Methods Monocytes obtained form human peripheral blood were induced into immature DCs and mature DCs in vitro. The whole antigen of hepatocarcinoma cells was extracted by freeze thawing method and used to sensitize immature DCs; Effective T cells were harvested by mixed lymphology reaction, and the specificity that effective T cells killed target hepatocarcinoma cells was examined by the MTT method. The effects of lycium bararum polysaccharides on DCs' maturity,T cell activation,proliferation and anti-tumor were studied. Results Lycium bararum can promote the maturity of DCs, DCs with whole antigen of hepatocarcinoma cells can activate specifically T cells into effective T cells which can kill hepato carcinoma cells specifically while not MG-63 cells. polysaccharides alone had no significant effect on the proliferation of T cells but they could promote the maturity of DCs and reinforce proliferation and anti-tumor effects of effective T cells. Conclusion Lycium bararum polysaccharides can promote the maturity of DCs and reinforce proliferation and anti-tumor effects of effective T cells.
