4-HPR促髓母细胞瘤细胞凋亡的机制
N-4-Hydroxyphenyl Retinoid-Induced Apoptosis is Associated with Down-Regulation of Bcl-xL in Human Medulloblastoma

摘要: 目的 探讨4-羟苯维胺酯对髓母细胞瘤细胞株的促凋亡的效应, 及其对抗凋亡相关基因Bcl-xL表达的影响。 方法 将2.5μM的4-羟苯维胺酯作用于髓母细胞瘤D283MED细胞株,用MTT实验,克隆形成实验检测4-羟苯维胺酯对细胞增殖的抑制作用,TUNEL法检测瘤细胞的凋亡率。采用RT-PCR及Western blot方法检测Bcl-xL的mRNA和蛋白的表达。 结果 4-羟苯维胺酯可显著抑制D283MED瘤细胞的增殖,明显促进瘤细胞凋亡(P<0.05);RT-PCR及Western blot结果显示Bcl-xL的mRNA和蛋白均明显减少。 结论 4-羟苯维胺酯通过下调Bcl-xL基因及其蛋白的表达,从而抑制髓母细胞瘤D283细胞的增殖,促进瘤细胞凋亡。Abstract: Objective To investigate the effects of N-4-hydroxyphenyl retinoid(4-HPR)-induced apoptosis and the expression of Bcl-xL in human medulloblastoma D283MED cell line. Methods Human medulloblastoma D283MED cell line was cultured in the medium containing 4-HPR( 2.5μM ) . The ability of ATRA to inhibit glioma cells proliferation was examined by cell viability analysis (MTT assay) and plate colony formation assay. The apoptosis was detected by TUNEL assay. The expression of Bcl-xL mRNA and protein in D283MED cells was detected by RT-PCR and Western blot respectively. Results 4-HPR inhibited cell proliferation and caused apoptosis in D283MED cell line. 4-HPR down-regulated the expression levels of Bcl-xL mRNA and protein. Conclusion 4-HPR might inhibit the proliferation of D283MED cells by down-regulating the expression of Bcl-xL.
