整合素在肺癌细胞中的差异表达

Analysis of Integrins Differential Expression in Lung Cancer Cells

  • 摘要: 目的 应用基因芯片技术研究整合素基因家族在三种肺癌细胞株A549(肺腺癌)、H1650(肺泡细胞癌)、DMS53(小细胞肺癌)中的表达差异。 方法 提取肺癌细胞A549、H1650、DMS53的mRNA,反转录为cDNA,以32P标记cDNA,与整合素基因芯片进行杂交,经放射自显影后读取灰度值并进行统计学分析。 结果 整合素基因家族中ITGB1在三种肺癌细胞A549、H1650和DMS53上均出现高表达;ITGA4在H1650的表达显著高于A549和DMS53 (>2倍);ITGA3在小细胞癌细胞DMS53中的表达显著低于非小细胞癌(NSCLC)细胞A549和H1650;ITGB6、ITGB8在DMS53中的表达显著高于A549。 结论 一些整合素亚单位在三种肺癌细胞株A549、H1650、DMS53中的表达谱存在差异,有望为肺癌的靶基因治疗提供实验依据。

     

    Abstract: Objective To analyze the expression of integrins in 3 different lung cancer cell lines including A549 (adenocarcinoma), H1650 (bronchioloalveolar carcinoma) and DMS53 (small-cell carcinoma). Methods The mRNA was extracted from A549, H1650 and DMS53, and then reversely transcribed to the cDNA. The mixed probes labeled by 32P were hybridized to the microarray chip, and exposed to a phosphor screen and scanned in a phosphoimager. Results ITGB1 integrin was observed with up-regulation in all the lung cancer cells. The expression level of several integrins including ITGA3, ITGA4, ITGB6 and ITGB8 displayed a>2.0-fold difference. Up expression of ITGA4 was detectable in H1650, and ITGA3 was observed in all the lung cancer cells except in DMS53, ITGB6 and ITGB8 were detectable in DMS53. Conclusion The expression of several integrins varied in three lung cancer cells. Integrins may play an important role in lung cancer metastasis.
