Abstract:
Objective To investigate the molecular mechanism on genomic level by using Fei-Yan-Ning Granulae in treating transplanted human lung carcinoma in nude mice. MethodsNude mice with transplanted human lung carcinoma H460 tumor were randomly divided into three groups:control group,chemotherapy group and Feiyanning Granulae treated group(FYN group).The total RNA were isolated from carcinoma tissue of groups,and the changes of genes expression profile of tumor tissues were evaluated by cDNA microarray technology. ResultsCompared with the control group,the chemotherapy group showed 2187 genes changes.The 997 genes were up-regulated and 1190 genes were down-regulated.The FYN group showed 2093 genes changes in their gene expression profiles.The 968 were up-regulated and 1025 gene were down-regulated,involving multiple scopes as cell cycle,signal tranduction,energy metabolism,protein synthesis,cell receptors as well. Conclusion The antitumor effect of Feiyanning granulae is multi-targeted,which elucidates part of the pharmacologic mechanism of Feiyanning granulae in molecular level.
