细胞周期及其相关蛋白在食管上皮癌变中的 表达及其意义
Expression of Cell Cycle Correlative Protein in Esophageal Cancer

摘要: 目的研究细胞周期调控因子细胞分裂周期素25A(cell division cycle 25 A,CDC25A )、转化生长因子-β(transforming growth factor-β, TGF-β)和Smad3蛋白在食管上皮癌变过程中的表达及其意义。方法利用流式细胞术检测38例同个体的食管癌、食管正常黏膜组织的细胞周期以及CDC25A、TGF-β、Smad3蛋白的表达。结果流式细胞术结果显示,食管癌与正常黏膜相比,增殖指数 (proliferation index,PI)显著增高(P<0.05),CDC25A蛋白显著增高(P<0.05),TGF-β、Smad3蛋白显著下降(P<0.05)。这些蛋白的表达量与年龄和性别无关(P>0.05),与组织学分级有关,低分化鳞癌与中高分化鳞癌相比,CDC25A蛋白显著增高(P<0.05),TGF-β、Smad3蛋白显著下降(P<0.05)。结论CDC25A、TGF-β、Smad3的表达失衡与食管癌的发生有密切关系,可作为食管癌的诊断指标。Abstract: To investigate the expression of cell cycle correlative protein CDC25A,TGF-β and Smad3 in esophageal carcinoma and the relationship between the abnormal expression of these factors and the development of esophageal carcinoma. Methods The expression of CDC25A,TGF-β and Smad3 protein and cell cycle of 38 esophageal carcinoma cases and normal esophageal mucosa at surgical margin of esophageal carcinoma tissues was detected by Flow cytometry(FCM). Results The expression of CDC25A in esophageal carcinoma was higher than normal tissue and the expression of TGF-β and Smad3 in esophageal carcinoma was lower than normal tissue. The expression of these proteins was related with tissue differentiation, but not age and gender. Conclusion The abnormal expression of CDC25A,TGF-β and Smad3 may be related with the origin of esophageal carcinoma, so it can be the diadynamic criterion of esophageal carcinoma.
