Abstract:
Objective To study how bcl-2 downregulation influence on proliferation, apoptosis, and invasion of glioma cell lines. Methods bcl-2 siRNA was chemically synthesized and transfected into glioma cell line SHG44 and TJ905. The silencing of bcl-2 expression in two cell lines were checked by Western blot.. The effect of bcl-2 supression on proliferation, invasion, and apoptosis of two cells were evaluated via MTT, cell transwell assay and Flow cytometric method. Results bcl-2 siRNA were effectively inhibited the expression of bcl-2 gene in SHG44 and TJ905 glioma cell lines. Silencing bcl-2 gene expression leads to the growth suppression of both SHG44 and TJ905 cells at different time point.Similarly, two cells invasion abilities were significantly reduced. However, no apparent effect on apoptosis was observed. Conclusion Our results have indicated that bcl-2 gene silencing effectively inhibited growth, proliferation and invasion of glioma cells.
