肝癌膜相关抗原HA b18G 的提纯及功能鉴定

Affinity Purification and Functional Identification of Hepatoma Membrane Associated Antigen HAb18G

摘要: 目的　从新鲜肝癌手术标本中提取肝癌膜相关抗原 HAb1 8G,并鉴定其诱导 MMPs分泌的功能。方法　用 HAb1 8m Ab制备亲和层析柱, 毛细管电泳分析纯化结果 ;酶谱法鉴定HAb1 8G的功能。结果　经亲和层析纯化后, HAb1 8G纯度达 96% ;酶谱法显示 HAb1 8G刺激人成纤维细胞分泌 MMPs的量及种类有明显变化。结论　亲和层析法获得了高纯度的肝癌膜相关抗原 HAb1 8G,其具有细胞外基质金属蛋白酶诱导因子 (即 EMMPRIN/CD1 47)的功能.

Abstract: Objective To botain hepatocarcinoma membrane antigen HAb18G from fresh surgical hepatoma specimen and identify its function. Methods HAb18G was obtained by affinity chromatograpic purification.The purity of HAb18G was test by high performonce capillary electrophoresis.Its function was identified by zymography.
