环孢素A对阿霉素治疗肝癌影响的实验研究

Influence of Combined Cyclosporine A with ADM on Hepatocellular Carcinoma

摘要: 目的探讨环孢素A(Cyclosporine A,CsA)对阿霉素(Adriamycin,ADM)治疗肝癌的影响。方法培养HepG-2肝癌细胞,分成对照、CsA、ADM、CsA+ADM组,分别处理24~48h,采用MTT法检测细胞增殖、流式细胞术测定细胞周期及细胞凋亡。结果肝癌细胞增殖能力48h明显受到抑制,CsA在5~1000μg/L随浓度增加,凋亡率逐渐增加,超过该浓度时,则出现凋亡率减低。CsA、及ADM均使G2期显著延长;CsA对HepG-2肝癌细胞有促进细胞凋亡作用,与ADM有协同促调亡作用。结论环孢素A对肝癌增殖有抑制作用,使G2期阻滞,促凋亡。环孢素A联合阿霉素对肝癌治疗有协同作用。

Abstract: Objective To investigate the influence of combined cyclosporine A with adriamycin on hepatocellular carcinoma. Methods HepG-2 cells were cultured,they were divided into 4 grops namely, control、CsA、ADM、CsA+ADM group, acted on respectively for 24 to 48 hours.
