槐耳颗粒联合沙利度胺抑制鼠肝癌H22细胞种植瘤的实验

Research of Huaier Granules Combined with Thalidomide Inhibits H22 Liver Cancer Cells Subcutaneously Implanted into Mice

  • 摘要: 目的 探讨槐耳颗粒和沙利度胺治疗肿瘤的作用机制。 方法 将雄性昆明小鼠(每组10只)分为对照组(A组)、槐耳颗粒组(B组)、沙利度胺组(C组)和槐耳颗粒+沙利度胺组(D组)。用H22细胞给小鼠皮下注射,复制皮下瘤模型。观察各组移植瘤质量和抑瘤率,采用免疫组织化学法检测各组瘤组织VEGF、CD31蛋白表达。 结果 与对照组比较,3个用药组小鼠的移植瘤重和瘤组织CD31阳性率显著低于对照组(P<0.01),槐耳颗粒+沙利度胺组VEGF表达显著低于对照组VEGF表达(P<0.01);与沙利度胺组和槐耳颗粒组比较,槐耳颗粒+沙利度胺组小鼠瘤质量和瘤组织CD31阳性率显著降低 (P<0.05)。 结论 槐耳颗粒和沙利度胺联合用药有明显抑制肿瘤生长的作用,其机制可能是通过下调VEGF蛋白表达和降低微血管密度,促进肿瘤细胞凋亡;并且两者有协同作用。

     

    Abstract: Objective To investigate the mechanism of Huaier Granules combined with Thalidomide inhibited tumor. Methods Male Kunming mice were randomly divided into controlled group(group A),Huaier granules group (group B), Thalidomide group(group C), and Huaier granules plus Thalidomide group(group D).Each group consisted of 10 mice. The tumor-bearing mice model was induced by subcutaneously injection of H22 cells.The mass of transplanted tumor and inhibition rate of tumor in all groups were observed.The expression of VEGF and CD31 protein was assayed by immunohistochemical method. Results Compared with those in control group,the mass of transplanted tumor tissue and the positive rate of CD31 protein in three treated groups were significantly decreased(P<0.01).And the positive rate of VEGF protein in group D was significantly decreased(P<0.01).Compared with those in group A and group B,the mass of tumor tissue and the positive rate of CD31 protein in group D were significantly decreased(P<0.05). Conclusion Huaier granules and Thalidomide had marked and synergy antitumor effect. The mechanism might be related to the apoptosis of tumor cells induced by the down-regulation of vascular endothelial growth factor(VEGF) protein.
