Expression of miRNA-200c in Gastric Cancer and Prediction of Its Target Gene RhoE

  • 摘要: 目的 研究miRNA-200c在胃癌和对应癌旁正常组织中的差异表达情况,探讨miRNA-200c与胃癌临床病理参数之间的相关性以及miRNA-200c在胃癌中可能调控的靶基因。方法 应用荧光定量-聚合酶链反应技术检测miRNA-200c在胃癌和癌旁正常组织的表达情况,利用生物信息学技术预测miRNA-200c可能的靶基因。结果 miRNA-200c在癌组织中的表达显著低于癌旁正常组织(P<0.05),miRNA-200c低表达与组织学分级呈显著相关,而与性别、年龄、肿瘤大小及TNM分期等无相关性。生物信息学预测RhoE可能是miRNA-200c的靶基因。结论 miRNA-200c在胃癌中低表达,与组织学分级呈相关性,miRNA-200c可能通过调控RhoE发挥生物学作用。

     

    Abstract: Objective To explore the expression of miRNA-200c in cancerous (T) and paired non-cancerous (N) tissues of gastric cancer,determine the correlation between miRNA-200c and clinicopathological characteristics and analyze the possible target genes. Methods Real-time PCR was used to quantify the expression of miRNA-200c.Bioinformatics was used to predict the target genes of miRNA-200c. Results miRNA-200c was significantly lower in cancerous tissues than that in the normal tissues (P<0.05).miRNA-200c was associated with histological grade.RhoE was the target gene of miRNA-200c. Conclusion Our results indicate that miRNA-200c plays an important role in gastric cancer and RhoE may serve as a target gene of miRNA-200c.
