Abstract:
Objective To explore the correlation between the level of IL-10 and TGF-β1 in portal vein and the ratio of Thl/Th2 like cytokines in liver previous to liver metastasis in mice models of metastasis. Methods Twenty mice were randomly assigned into control group and experimental group.Established the orthotopic model of liver metastasis by implanting tumor block into the cecal wall in BALB/c mice as experimental group,and the other ten were accepted the sham operation.After two weeks,all mice were killed and blood samples in portal veins and liver samples were collected for ELISA testing of IL-10,TGF-β1 and IFN-γ.The t-test was used to compare the differences between two groups,and Bivariate correlation analysis was used to analyze the correlation. Results Compared to the control group,the ratio of Th1/Th2(IFN-γ/IL-10) in livers of experimental mice was decreased significantly,which showed a significantly negative correlation with serum levels of IL-10 and TGF-β1 in portal vein. Conclusion Tumors of mouse colon can produce and release IL-10 and TGF-β1 into portal vein,which can flow to liver and lead to the shift of Th1/Th2 like cytokines to promote the liver metastasis.
