Abstract:
ObjectiveTo study the effect of the EGFRvⅢ siRNA on the expression of EGFRvⅢ and the apoptosis and cell growth in the U87-EGFRvⅢ cell line. Methods The EGFRvⅢ siRNA was synthesized by the chemical method.The gene expression of EGFRvⅢ was detected by RT-PCR and the protein expression was detected by Western blot and immunofluorescence.MTT and Annexin V-FITC/PI assays were used to detect the inhibitory effect on cell proliferation and the apoptosis effect. Results The results of RT-PCR and Western blot showed that the gene and protein expression of EGFRvⅢ was decreased significantly by EGFRvⅢ siRNA,which could be reduced (75.7±3.1)% after transfected with siRNA for 48 h.The results of the MTT and Annexin V-FITC/PI assays showed that siRNA could suppress the cell growth of glioma cell line and induce apoptosis of the glioma cell. ConclusionEGFRvⅢ siRNA could significantly inhibit EGFRvⅢ gene,protein expression and the proliferation,as well as induced apoptosis of U87-EGFRvⅢ cells.
