摘要: 目的探讨RNA干扰技术沉默survivin基因对口腔表皮样癌细胞株KB增殖和凋亡的影响。方法化学合成靶向survivin基因的小干扰RNA片段(siRNA),脂质体法转染KB细胞。通过RT-PCR、蛋白印迹法分别检测survivin mRNA和蛋白表达变化,流式细胞仪检测细胞生长周期和凋亡,MTT法检测转染后KB细胞增殖情况。结果siRNA可以有效地降低KB细胞survivin mRNA和蛋白的表达;转染后KB细胞阻滞于G2/M期,凋亡增加,增殖受抑制。结论应用RNA干扰技术沉默survivin 在KB细胞中的表达可以有效抑制该细胞增殖、促进癌细胞凋亡,将survivin作为口腔癌基因治疗的靶点,通过RNA干扰技术对口腔癌进行基因治疗具有一定的可行性。Abstract: ObjectiveTo observe the inhibitory effect on oral cancer cell lines KB induced by RNA interference (RNAi) of survivin gene. MethodsChemically synthesized small interference RNA(siRNA) targeting survivin was transfected into KB cells by lipofectamine method. The expression of survivin mRNA and protein were detected by RT- PCR and Western blot respectively. The cell cycle and apoptosis were detected by flow cytometry(FCM).The growth of KB cells was detected by MTT. ResultsThe expression of survivin mRNA and protein were obviously reduced. The cell cycle of KB cells was arrested at G2/M phase.The apoptosis rate was increased and the growth of cells was inhibited. Conclusionsurvivin siRNA can induce apoptosis and reduce proliferation of KB cells by silencing survivin gene expression. RNAi targeting survivin has a potential value in gene therapy of oral cancer.
