乳腺癌组织和癌旁组织MGMT和GST-π蛋白表达的流式细胞术分析

摘要: 目的研究乳腺癌组织和癌旁组织O6甲基鸟嘌呤DNA甲基转移酶(MGMT)和谷胱苷肽S转移酶π(GSTπ)蛋白表达的临床意义。方法应用流式细胞术(FCM)分析MGMT和GSTπ蛋白表达。结果乳腺癌组织MGMT和GSTπ表达显著高于癌旁组织(P<0.05)。2种耐药相关蛋白在不同年龄、不同肿瘤大小、不同病理类型、不同临床分期和雌、孕激素受体阴性与阳性患者间的表达均无显著差异(P>0.05)。有淋巴结转移组患者MGMT与无淋巴结转移组患者比较无显著差异(P>0.05)。但GSTπ表达在有淋巴结转移组患者显著高于无淋巴结转移组患者(P<0.05)。相关分析显示,MGMT与GSTπ表达间无显著相关性(P>0.05)。结论初步研究结果提示,MGMT蛋白表达可能与乳腺癌预后无关,不能作为乳腺癌的一个预后指标。GSTπ表达与淋巴结转移状况有关,有可能成为预测乳腺癌预后的指标之一。Abstract: Objective Our purpose was to explore the clinical significance of MGMT and GST-π protein expression in breast carcinoma tissues and corresponding para-carcinoma tissues. Methods Flow cytometry was used to analyze the expression of MGMT and GST-π protein.
