用免疫组化和原位杂交方法检测和分析人骨肉瘤组织中Ⅰ、Ⅱ和Ⅲ 型胶原蛋白和mRNA 表达

摘要: 目的 观察骨肉瘤组织 、 和 型胶原蛋白和 m RNA的表达及其与骨肉瘤分型和分化的关系。方法 免疫组化 LSAB法检测胶原蛋白, 原位杂交检测胶原 m RNA的表达, 用图像分析系统进行分析和半定量测定。结果 骨肉瘤组织不仅表达 型胶原蛋白 (阳性率 87%, 灰度值1 71 .99± 1 4.74), 并出现了 型和 型胶原蛋白 (阳性率分别为 30 .4%和 43.4%, 灰度值分别为1 5 3.0 7± 1 8.82和 1 68.2 9± 1 8.36) ;软骨母细胞型和混合型骨肉瘤 型胶原的表达高于其它型, 差异有显著性 ( P<0 .0 1 ) ;高中低分化的三组骨肉瘤间, 型胶原表达差异有显著性 ( P<0 .0 1 ), 分化越低表达越少。 、 型胶原 m RNA表达与蛋白表达所得结论基本一致。结论 型胶原可作为软骨母细胞型和混合型骨肉瘤的分型参考指标, 型胶原可作为骨肉瘤分化及恶性度判断的参考指标.Abstract: Objective To investigate expression of type Ⅰ?Ⅱ and Ⅲ collagen in human osteosarcoma and their relationship with classification and differentiation of osteosarcoma.
